Medetomidine HCl is a selective α2-adrenoceptor (α2-adrenoceptor) agonist with a Ki of 1.08 nM, which is 1620-fold more selective than acting on α1-adrenoceptor.

Medetomidine is a selective α2-adrenal receptor agonist with a Ki of 1.08 nM, 1620-fold selectivity over α1-adrenal receptors, and has little or no binding to other neurotransmitter receptors.

In anesthetized rats, medetomidine (1-100 μg/kg, intravenously) causes a dose-dependent transient decrease in blood pressure and heart rate. In brain-destroying spinal cord rats, medetomidine showed potent vasopressor (PD501.7 μg/kg) and sympathetic inhibition (ID501.6 μg/kg) without affecting the basal heart rate. Medetomidine causes a dose-dependent sedative effect, at high doses (>100 mcg / kg), causes a loss of positive reflexes, and lowers body temperature. Medetomidine causes a decrease in biogenic amine turnover in the brain, dose-dependent inhibition of norepinephrine (NE) inversion, inhibition of cerebral dopamine turnover at high doses, and decrease of serotonin reversal.
