Levodropropizine (Levodropropizine) is a chiral antitussive drug first launched by Dompe'Farm company in Italy. The drug inhibits the cough reflex by acting on C-fiber in the respiratory tract. It is clinically used for the treatment of intense phlegm-free dry cough. It has almost no effect on the central system, good selectivity, strong antitussive effect, and no lethargy, respiratory depression, dependence and other side effects, compared with racemic, it has less side effects.

 

Pharmacological action
This product is a peripheral antitussive drug, which plays a antitussive role by selectively inhibiting the peripheral C-fibers of trachea and bronchus. It has a strong antitussive effect and can be maintained for a long time. Because it does not interact with β adrenal receptors, M choline receptors, and opioid receptors, it has fewer adverse effects of central inhibition. Subacute and long-term toxicity tests in rats and dogs showed that the main symptoms during the treatment period were salivation, decreased intake and body weight in rats, and increased central sedation, peripheral vasodilation and heart rate in dogs. Hepatotoxicity in high doses to both animals.

In vivo study
In rats, Levodropropizine has a weaker central sedative effect than racemic tartrate, and it does not cause physical dependence. Levodropropizine(15 mg/kg, intravenously) simultaneously reduces the duration of asphyxia and the C-fiber response to phenylformin. Lvdp-induced inhibition of C-fiber response to PBG averaged 50% in lung fibers and 25% in non-lung fibers. Levodropropizine has been shown to have good antitussive activity in anesthetized guinea pigs and rabbits. Levodropropizine(200 mg/kg) inhibits substance P extravasation, but it does not affect platelet activating factor induced extravasation.
