Palonosetron hydrochloride, an anti-nausea and vomiting drug, is a novel high-selectivity and high-affinity 5-HT3 receptor antagonist, which blocks the presynaptic 5-HT of central peripheral neurons of the gag reflex. receptors excitate and directly affect the central nervous system within 5-HT. The impulse generated by receptor excitation acts through the vagus nerve to the posterior area, blocking the vagus nerve endings in the intestine and preventing the signal from being transmitted to 5-HT. receptor trigger zone, which reduces the incidence of nausea and vomiting. It is clinically used to treat acute and delayed nausea and vomiting caused by moderate and severe emetogenic chemotherapy drugs. Chemicalbook has attracted much attention because of its high efficacy, low toxicity and side effects, long half-life (about 40h), and small dosage. Palonosetron hydrochloride was prepared by splitting, amination, reduction, ring closure and salt formation with 1,2,3,4-tetrahydro-1-naphthalenecarboxylic acid as the starting material. Clinical studies have shown that palonosetron is safe with corticosteroids, analgesics, antiemetics, antispasmodics, and anticholinergics. Studies in murine tumor models have shown that palonosetron does not inhibit the anticancer activity of the five chemotherapeutic agents studied (cisplatin, cyclophosphamide, cytarabine, doxorubicin, and mitomycin C).
